甘蓝夜蛾核多角体病毒活体增殖因子及最佳增殖条件的研究

Influencing Factor and Optimal Conditions for in vivo Production of MabrNPV

摘要: 甘蓝夜蛾核多角体病毒Mamestra brassicae nucleopolyhedrovirus(简称MabrNPV)是甘蓝夜蛾最重要的昆虫病原微生物。本文采用正交法研究影响MabrNPV在甘蓝夜蛾体内增殖的主要因子及因子的主次顺序,确定互作因子并筛选病毒增殖最佳条件。研究结果表明,影响MabrNPV产量的因子及互作因子主次顺序为C(饲毒虫日龄) > C×D(饲毒虫日龄与收毒时间互作) > B×C(饲毒剂量与饲毒虫日龄互作) > B×D(饲毒剂量与收毒时间互作) > B(饲毒剂量) > D(收毒时间) > A(温度)。明确因子C对病毒产量有极显著影响,因子C和D存在互作且对产量有显著影响。确定MabrNPV增殖生产的最佳条件为25 ℃下、以500 PIB/虫的剂量饲喂12日龄幼虫,9 d后提取病毒,平均单虫病毒产量为1.921×109 PIB,幼虫孵化到收集幼虫提取病毒的生产周期为21 d,病毒增殖3.842×106倍。

Abstract: Mamestra brassicae nucleopolyhedrovirus (MabrNPV) is the most important pathogenic microorganisms against cabbage moth. Major factors influencing mass multiplication of MabrNPV in vivo in the cabbage moth larvae and the role of these factors were studied by the orthogonal method. Factor interactions and the optimal conditions for mass production of MabrNPV were also investigated. The factors influencing MabrNPV yield were ranked according to their significance: larval age at virus treatment, interaction between larval age at virus treatment and virus harvest time, interaction between virus dosage and larval age at virus treatment, interaction between virus dosage and virus harvest time, virus dosage, virus harvest time, and temperature. Larval age at virus treatment and its interaction with virus harvest time significantly affected the production of MabrNPV. The optimal production conditions were: harvest virus at the 9th d post treatment of 12 d old larvae with 5×102 polyhedral inclusion bodies (PIB) per larva at 25 ℃. Under these conditions, MabrNPV production was 1.921×109 PIB per inoculated larvae. The whole production cycle of MabrNPV lasted 21 d starting from larvae hatching to virus harvest. The reproduction rate of PIB was 3.842×106 times.
